Spectrophotometric: C = A260 × 50 ng/μL (for dsDNA). Molecular weight: MW = bp × 660 g/mol. Copy number (molecules/μL) = (C × 10⁻⁹ / MW) × 6.022×10²³. Total mass (ng) = (copies × MW / Nₐ) × 10⁹.
